Description

Intended Uses: The AMH ELISA is an enzyme immunoassay for the measurement of Anti-Müllerian Hormone (AMH) in serum or plasma (EDTA or lithium heparin plasma). For research use only -

Principle of the Assay: The AMH ELISA is a solid phase enzyme-linked immunosorbent assay (ELISA) based on the sandwich principle. The microtiter wells are coated with a monoclonal (mouse) antibody directed towards a unique antigenic site of the AMH molecule. During the first incubation, the AMH in the added sample bind to the immobilized antibody. The simultaneously added enzyme conjugate, which contains an AMH antibody conjugated to horseradish peroxidase, binds to the AMH forming a sandwich complex. After a washing step to remove all unbound substances, the solid phase is incubated with the substrate solution. The colorimetric reaction is stopped by addition of stop solution, and optical density (OD) of the resulting yellow product is measured. The intensity of colour is proportional to the concentration of the analyte in the sample. A standard curve is constructed by plotting OD values against concentrations of standards, and concentrations of unknown samples are determined using this standard curve
